Vedr. Peer Reviewet fra den 14. juni omhandlende 2-73 og 3-71, fandt jeg denne analyse på IHUB fra en ellers altid faktuel baseret dygtig skribent gennem + 6 år.
Hvis analytiker fra større investeringsselskaber og andre institutioner kommer frem til en tilsvarende konklusion af Peer Reviewet og casen Anavex - så kan det nok forklare den seneste kursudvikling og øget volumen i handlede aktier.14 June 2021 Press Release Commentary
Today's press release ties together multiple scientific hypotheses surrounding Blarcamesine (Anavex 2-73) and its seemingly endless potential.
Over the years, Anavex has used a robust compilation of preclinical and clinical data to lead investors through an analytic maze of therapeutic possibilities and pathways. While the S1R agonist efficacy and safety are definitively evident, investors and scientists have been left stringing together the pieces as to "how" and "why". Finally, on 14 June 2021, "The emerging role of the sigma-1 receptor in autophagy: Hand-in-hand targets for the treatment of Alzheimer's" was published, connecting most of these dots.
To begin, in normal (unhindered) adults, S1R expression increases with age, with the opposite being true in Alzheimer patients. The paper continues to state that S1R is likely triggered in old age when other key receptors (M1/M4 muscarinic, D1/D2 dopamine, and 5HT2A serotonin) begin to fail. In working cells, the S1R would act as a fail-safe, and correct the deficiencies of the other receptors. Through autophagy, the cell would return to homeostasis. However, in Alzheimer patients, autophagic function and S1R expression deteriorate, leading to catastrophic failure of the cell.
In the case of Blarcamesine, S1R is activated, allowing for autophagic function to continue, and the return of homeostasis - ultimately saving and repairing damaged cells. S1R modulates glutamate levels, maintains endoplasmic reticulum, promotes neurogenesis, and suppresses neuroinflammation, in addition to other effects noted later. Most critically, these effects have recently been proven in PET imaging of Alzheimer patients. Of interest, the scientific community can be assuaged knowing the S1R promotes not only autophagy, but also inhibits amyloid beta. Of interest to Anavex shareholders, Biogen's Aducanumab which was just approved by the FDA sought (and failed) to demonstrate that removing amyloid beta resulted in better clinical outcomes. To date, Anavex has proven that amyloid beta is not the cause of Alzheimer's, but a byproduct. Due to this, the scientific community (still latching unto amyloid hypothesis) should be pleased with Blarcamesine' amyloid clearing, even though amyloid does not cause patient symptoms or degradation.
Alzheimer's is multifactorial, which is why S1R agonists are likely a potent strategy at addressing symptoms and reversing pathology. To comment further on regulating dopamine/serotonin, in-patient trials with Blarcamesine has revealed significant betterment for patient daily living to include increased REM sleep and better mood. These effects have been noted by patients and caregivers alike. This aligns very well with the PET imaging - solidifying the link between these tangible benefits and return to cellular homeostasis at the molecular level.
To date, Anavex' small 2a Alzheimer study has produced the most impressive cognitive data over any competitor. With an impressive safety profile and numerous daily living benefits, patients enjoyed 9% better cognitive scores over baseline at week 57, and 14% better cognitive scores over baseline at week 70. To be clear, some of these patients reversed decline. This Alzheimer data was correlated to a larger, phenomenally successful Parkinson's Disease Dementia trial. Only Cassava Sciences has published data that comes even close to Anavex data, and even then, the cognitive results were not quite as robust.
Perhaps the most titillating excerpt from the paper is as follows: "in the future it may be the case that SIGMAR1 ligands (or drug combinations) targeting the activation of autophagic, and other SIGMAR1 related neuroprotective pathways, are prescribed prophylactically, in much the same way as with statins for the preventative of heart disease today (aspirin) in an effort to prevent the loss of the SIGMAR1 receptor seen during AD".
To summarize, the newly published PET imaging results, combined with preclinical and clinical Blarcamesine data lends great credence to the probability of Anavex holding a miracle drug capable of diffusing many central nervous system (and other) disorders. With this in mind, and combined with the potential for outright prevention, Anavex has a high chance of successfully marketing their drugs (2-73 and 3-71). Revenues earned by such a platform would certainly produce a top-tier biopharma with a market cap in the 100's of billions.

En sådan forholdsvis stigning har vi, trods en pæn opadgående kurve, ikke set helt i samme grad i Anavex endnu.Mit pejlemærke er samme MC i Anavex, som for SAVA - hvilket ca. svarer til kurs 50 $.
Omvendt, hvis vi skal derop, kræver det min. gode Peer Reviews - og er de rigtig overbevisende burde vi også overgå SAVA betydelig.
Når vi de 50 $ tager jeg også 10-20 % ud og reinvester i mere "sikre" aktier - f.eks. DSV, Mærsk,Novo Nordisk, Genmab m.m.
Men alt står og falder med disse ventede Peer Reviews - leverer Anavex ikke på dette, skal vi være glade for en kurs på plus 10 $.

Det boer bemærkes at det forsøg var gjort med den lille dose på 5mg 2-73 en gang per dag. Saa det må jo give endnu bedre resultater i større doser.Godt gang i den i pre-market, stor volumen.

Bekræfter samtidig at MOA højst sandsynlig kan omsættes på tilsvarende måde fra præklinisk til klinisk - i andre indikationer indikationer!
Man skal godt nok lede længe efter noget som helst negativt i denne meddelse - og det bedste er, at dette ikke en gang er selve Peer Reviewet.
Understøtter denne disse data som forventet, burde vi få endnu et boost op!Samme strategi for Parkinson i næste uge Missling - tak!

Solsen, Tasso . Er der mon nogen, der ved hvor udbredt gruppen med denne Sigma variant er? Er det ikke væsentligt at vide?. Havde alle i dette Rett forsøg denne Sigma variant, hvor der altså , som jeg forstår det, kunne ses god effekt.
Med hensyn til PDD trial forestiller jeg mig også, at vi på et tidspunkt får at vide, at stoffet virker på en gruppe med en kendt biomarkør? Om det drejer sig om en stor eller lille andel af PDD - patienterne er vel afgørende for hvor stor succes stoffet får. Eller kender vi allerede denne gruppes størrelse?
Måske har TASSO forklaret det tidligere? Jeg forventer ikke at kunne forstå det hele, men det der med biomarkører spekulerer jeg en del over. -

Første af nok et øget og antal opgraderinger, samt tilgang af nye analytikere og deres kursmål.
For hvem med respekt for sig selv, kan udlade at dække et selskab, som måske kommer med det første virkelig gennembrud inden for CNS???
F.eks. burde Black Rock med deres 4.300.000 aktier melde ud, hvor de ser Anavex henne om et års tid.Tænker lige som Solsen, at det er de godt 80 % WT, som gælder alle mennesker og er bedst disponeret for en god effekt med 2-73, der henvises til. Mener dog også at der specifikt for RETT patienter var forskel på noget der hedder APOE gen - har dog ikke lige haft tid til at læse efter endnu.

når man kigger på sava og Annovis Bio, Inc. (ANVS), forståer jeg ikke helt hvorfor anavex ikke også er oppe på 100$ lige som de to andre.
hvis deres produkt er lige så godt eller bedre. er der nogle der kan forklarer det.
men ellers er det ikke så ringe end dag ,men det kunne værer bedre -
Ting tager tid. Men Anavex burde ligge med højere Mcap end dem du nævner.
Om de så er kontrakt værdisat er en anden sag.
Dagens data er efter min mening ikke belønnet nok. Men har sikkert noget med, at det er Rett indikationen.
Men kommer tilsvarende overbevisende data i PDD så er vi på $3bn Mcap.

Du kan ikke umiddelbart sammenligne kurserne på 2 forskellige aktier UDEN at tage antal cirkulerede aktier i betragtning. Hvis du vil sammenligne bør du i stedet se på selskabernes markedsværdier. Om det så giver et rigtigt billede af selskabernes reelle værdi, vil kun fremtiden kunne vise. Pt. mener "markedet" at SAVA' værdi er ca.
$ 3,5 mia, AVXL ca. $ 1,8 mia. og ANVS ca. $ 0,66 mia, se nedenstående oversigt

Markant og bred god respons i ældre piger, som ellers burde være næsten umuligt at se nogen forbedring i - og så endda på lav dosis i kun 7 uger.
Signifikant respons allerede efter 4 uger og effekten er endda tiltaget efter 7 uger.Dette tegner godt for forsøget med ældre piger på høj dosis senere i år - og endnu bedre for forsøget med yngre piger på høj dosis og 12 ugers varighed!
Resultater i extension forsøgene må blive virkelige gode.
Alt dette uden bivirkninger og med meget lille usikkerhed.
Sandsynliggør at precision tilgangen kan overføres til andre indikationer!
FDA kan ikke undgå at godkende 2-73 - måske allerede i år via. acc. godkendelse.

Efter dette kan 2-73 i praksis udskrives i andre indikationer også - udløser desuden en voucher til en værdi af + 100 mill. $!Ganske få kender til RETT - men når Anavex inden 1. juli udgiver tilsvarende eller efter min overbevisning endnu bedre resultater i PDD, så får vi en helt anden markant respons i aktiekursen!
70 % af målepunkterne i PDD har sammenfald med Alzheimer - når dette går op for markedet for alvor, tør jeg ikke spå om kursen!Rigtig mange anede end ikke om eksistensen af Anavex for bare få uger siden.
