    Are We Closer Than Ever to a Biotech Bottom?

    Some observations point to hope for an upturn.

    As we live through the steepest and longest bear market in biotech history, historical precedents offer little value in assessing how much lower we could fall or for how long. According to a recent biotech market update from Tim Opler and team at Torreya, over one-third of biotechs have lost 75% of their value since the market peaked in early 2021, over 200 companies are trading below enterprise value, and two-thirds of biotechs have an enterprise value below $100 million (vs. 36% in November 2021).

    That said, it is likely true that we may be closer than ever to a bottom. In fact, the week of May 16 was the first up week for the sector in the seven weeks prior-shockingly, it was actually the top-performing sector.

    Perhaps, this may have been ushered in part by the increasingly pervasive fears about an impending recession fed by disappointing earnings reports from leading consumer and retail companies flooded with excess inventory and facing soaring costs. Apparently, customers are trading down to cheaper private-label items and appear to be buying what they "need" vs. what they "want," and the inventory bulge is in the products that fall into the latter category. Inventory markdown risks are running high.

    Giants like Walmart and Target have seen their worst stock declines since 1987 and took a sharp ax to their forecasts. Investors fear it could get worse, as the likes of Costco, Dollar General, and Best Buy report their results.

    A rapid acceleration in the economy had investors rotating out of expensive growth stocks (like biotech), while rising expectations for rates penalized long-dated assets, such as the highly risky development-stage biotechs, years away from the market, and serial capital raisers highly dependent on capital markets. With the sputtering in the economy and prevailing expectations for a recession, investors may rotate back into growth, including biotech.

    Green shoots suggest optimism for biotech
    While still slow and selective, a window into accessing the capital markets does appear to be opening with IPOs and follow-on offerings picking up. We are hearing that the sector specialists do believe there is opportunity in the carnage, and money appears to be coalescing with a goal of identifying and capitalizing on them. Restructurings are reducing cash burns and ring-fencing lead programs. Firepower among the large-cap biopharmas, according to a report published by EY, is at all-time highs, and M&A, partnerships, and alliances remain a key pillar of growth strategies, particularly as the industry heads into a substantial patent cliff and industry blockbusters face losses of exclusivity. New funds continue to pour into the sector earmarked for investments in the innovation renaissance underway. ATLAS is closing a $3 billion fund, Sofinnova is bringing in $1.5 billion from Apollo, along with other VCs and financial sponsors with rising interest in the sector. Flagship and many others have all raised record amounts of capital for investment in biotech.

    Smart money looks to invest more in biotech
    "This is not 2000, when biotech investors often bet on a one-product company, which by definition is binary. Today, savvy healthcare investors have an opportunity to buy platforms of IP, with multiple paths to value creation, some of which have the potential to provide cures," says Meghan Fitzgerald, a private equity investor who is also a founder in K2Healthcare Ventures and an active life sciences fund investor. "For those with the right teams to assess the science, it's a compelling time to roll up assets trading at or below cash."

    Interest in reverse mergers is also growing, which have the potential to create new companies with better funding and broader pipelines. Reverse mergers could consolidate the public biotech space. According to select equity capital market desks, dialogues with prominent funds in the sector are calling for a bottom. Let's hope they are right.

      75 % rammer vist meget godt nedturen i min portefølje. Det er da helt vildt det her. Så længe det fundamentale er intakt må man bare bevare roen, holde isen kold. Mon ikke det retter sig. Spændende om jeg skal pensioneres i 2027 eller 2030. Hvis dette fortsætter truer en forlængelse af kontrakten, uha, en grim tanke.

          Det er da en fantastisk artikel Makingmoney.Så venter vi blot med spænding på de sidste resultater. Jeg tror det bliver rigtig stort, og vil gøre rigtig mange menneskers liv meget bedre.

            Virker som en seriøs og umiddelbar kompetent tilgang.
            Selvom de selv har investeret i Anavex, synes de at have styr på argumenterne, hvorfor Anavex adskiller sig fra øvrige tilgange på området.

Our modeling studies suggest that Anavex2-73 is unique in that it could also be a potent FKBP12 inhibitor, even Anavex (the company) is not aware of it! Most likely Anavex2-73 will succeed in clinical trial in AD because it works in both pathways!)

            Have we finally found a compound that can do both, activate S1R and inhibit FKBP12? Is there light at the end of the tunnel in AD drug development? All of us are waiting with abated breath for the trial results from Anavex. Maybe, just maybe, we can relax knowing the mechanisms of Anavex2-73. The possible dual actions of Anavex2-73 might project altered course of disease progression as well as improved memory and cognition for AD patients.

            (Lay Summary: Upon laboratory confirmation of the modeling studies, Anavex2-73 could change the way we treat or see Alzheimer's disease. It could restore memory function and completely change the trajectory of disease progression. As a matter of fact, it should work on other diseases as well such as Parkinson's, ALS, Fragile X syndrome, Rett, and possibly others. As for the ongoing trials, the results could be so spectacular that we would have never expected!)

              Han er gået videnskabligt til sagen med en imponerende referenceliste !

              Vi er tæt på R&D dagennså spændingen stiger om end vi nok måske må nøjes med det videnskablige samt lidt analyser. Sikkert ikke meget om deres forretningsmæssige syn på fremtiden.

              Så for nogle dage siden på Ihub en interessant ansættelse af en erfaren NDA (erfaring med filing af resultater til myndighederne) mand 🙂

              En søgning på LikedIn viser også der nok er ansat en del medarbejdere i Anavex. Så håb om at de har lidt flere ressoucer fremover, hvilket er nødvendigt for at få lidt speed på tingene.

              Jeg har også hørt på Rett webcastet fra 18. maj. Meget opmuntrene at gøre at en 41 årig Rett pts for første gang siden hun var tre år kunne noget vokalt. En anden pts kunne udtale investigatorens navn, hvilket han ikke forventede ved forsøgets start.

              Kaufmann fra Anavex oplyste, at fda var presset på bemanding, hvorfor alt med dem tog dobbelt så meget tid end sædvanligt. Et forhold der delvis forklarer, hvorfor vi synes det går for langsomt.

              Lad os håbe, at det lykkes i CNS sygdommene for Anavex. Senest til morgen har jeg læst at endnu et forsøg i Alzheimers er fejlet (Genentech).

                      Ikke rart for de hårdtprøvet og tålmodige aktionærer, at se en volume på 200.000 kan få kursen ned med 10% og indsidder samtidig sælger fra ( måske han er blevet anbefalet af sælge i dette bearmarked ) dårligt signal

                      Men som skrevet før, den kan falde 10 den ene dag og 10 den næste dag, så derfor er jeg også meget forsigtig med mine køb/ det samme gælder i andre suppleringer

                      Men hen over tid, har market det med at stiger, der er dog meget geopolitisk uro i øjeblikket- inflation- krig stigende forbrugerpriser- energipriser- og jokeren coronavarianter mm.

                        Anavex ... Utroligt at de ikke er kommet med et program for R&D dagen den 21. juni ???

                        Mht insidere udnyttede Missling den 29. Juni sidste år sine warrants (50.000 stk) og solgte aktierne for $26.

                        Det kunne være interessant, hvis han kunne gentage bedriften i år 🙂

                          Et par af de årsager jeg især også kan tilslutte mig:

                          Især begyndende opkøb fra BP indikere, at der er værdier i biotek selskaber, der overstiger nuværende kursleje.

                          Biotek er ikke cykliske selskaber, men skal værdisættes efter deres videnskabelig resultater/værdier - biotek har dog trods dette taget turen ned med det øvrige marked.

                          Der er et relativt positivt nyhedsflow fra sektoren og især shorterne vil komme i klemme, nå de kommer fra kunstig pressede biotek selskaber - shorterne slipper deres tag i aktien og går videre.

                          FDA er måske nu mindre overbebyrdet af COVID-19 relaterede ansøgninger mm. og begynder at kunne fokusere mere på normal situationen og behandlingstider. Ved sidste CC nævnte Anavex, at hele proceduren med FDA var trukket meget ud netop pga. af COVID-19.

                          Sammen med det historiske markante fald i bl.a XBI-indekset til et relativt meget lavt niveau, har man tidligere set en trendvending fra netop dette kursleje.

                          Uanset geopolitiske og markedsøkonomiske forholde, vil der altid være et behov for ny og bedre behandlingsmuligheder - ja måske endnu mere nu, da en bedre medicin måske ville kunne være med til at aflaste pleje/sundhedssektoren, som jeg er overbevist, at bl.a Anavex ville kunne bidrage med i form af 2-73 of 3-71.

                          Solsen - ja meget underligt, at Anavex ikke kommer med mere vedr. R & D dagen - især da Missling gjorde så meget ud af begivenheden på sidste CC?
                          Håber lidt naivt på, at de lige kan nå at få nogle data fra PD OLE forsøget med, som burde har haft deres sidste patient igennem de 48 uger i april 2022?

                          Vi kan kun afvente med det i mente, at data/videnskaben hidtil kun har været positiv - der har absolut intet negativt været fra hverken prækliniske eller kliniske forsøg!
                          Uden nogen som helst form for seriøse bivirkninger!

                          PS: Anavex er tilmeldt AAIC den 2. aug. 2022:

                          Der er også et nyere abstrakt tilgænglig vedr. 3-71 i Alzheimer ( jeg har ikke set den før ), som er særdeles positive:
                          Selv efter udvaskning efter 3-71, bibeholdes den meget positive cognitive effekt.
                          Der nævnes også, at 3-71 sandsynligvis vil have en præventiv effekt mod udvikling af cognitive indikation/ Alzheimer.

                          Chiara Orciani, MSc; Sonia Do Carmo, PhD; Morgan K Foret, PhD; Hélène Hall, PharmD, PhD; Quentin Bonomo, BSc; Agustina Lavagna, MSc; Chunwei Huang, MD and Claudio A Cuello, MD, McGill University, Montreal, QC, Canada

                          Background: Acetylcholinesterase inhibitors represent four out of six drugs for treating Alzheimer's disease (AD), with time-limited efficacy likely due to the progressive loss of cholinergic neurons. The selective allosteric M1 muscarinic and sigma-1 receptor agonist Anavex 3-71 (aka AF710B) [1] takes advantage of the fact that acetylcholine postsynaptic M1 muscarinic brain receptor levels remain unchanged in AD. Previously, Anavex 3-71 treatment attenuated AD hallmarks in McGill-R-Thy1-APP transgenic rats when administered at advanced stages of the AD-like pathology [2]. However, preventive strategies instead of late treatment should be more effective in AD. Therefore, we tested whether Anavex 3-71 administration during early amyloid pathology stages could prevent cognitive impairment, plaque deposition and neuroinflammation.

                          Method: Pre-plaque, seven-month-old McGill-R-Thy1-APP rats were subjected to daily oral administration of Anavex 3-71 (10 µg/kg) for seven months. After one month of drug interruption (wash-out), we performed Novel Object Recognition (NOR), Morris Water Maze (MWM) and Social Preference (SP) behavioural tests. Subsequently, brains were extracted, fixed and analyzed by histochemistry and immunohistochemistry.

                          Result: McGill-APP rats had deficits in discriminating the novel object in the NOR and locating the hidden platform in the MWM compared to wild-type littermates. Anavex 3-71 restored the abilities of McGill-APP rats in the NOR task to wild-type levels and partially rescued the deficits in the MWM. In the SP task, Anavex 3-71-treated McGill-APP rats exhibited diminished deficits in SP interaction time with a "stranger rat". Treatment significantly reduced the number of mature Thioflavin-S positive plaques in the subiculum compared to vehicle-treated McGill-APP rats and diminished the number of diffuse plaques in the cortex and hippocampus. Furthermore, Anavex 3-71 treatment significantly reduced microglia and astrocyte recruitment towards CA1 hippocampal Aß-burdened neurons compared to untreated McGill-APP rats.

                          Conclusion: The long-lasting effect of Anavex 3-71 in preventing cognitive decline of McGill-R-Thy1-APP rats, even after a wash-out period would suggest some preventative, disease-modifying, properties of the compound over the AD-like amyloid pathology. This is supported by the attenuation of extracellular Aß deposition and reduced glial cell recruitment towards hippocampal neurons. [1] Fisher et al., 2016; Neurodegenerative Diseases.                                 Vel ikke noget revolutionerende nyt.

                                Anavex poisoned for significant success var et af udsagnene 🙂

                                Et andet at patienter vil kunne undgå alzheimers, parkinson, demens ets ved profylaktisk at indtage Blarcamesine/2-73

                                Flere ansatte deltog og var overbevisende.

                                Ikke sikkert det rykker på kursen.

                                Min tid tillader desværre ikke mere lige nu. Men mon ikke nogle mere kloge end jeg har noget mere end dette.

                                      Det som jeg tænker er, at det er en salgsvideo til big pharma selskaber. Man gør sig lækker. Hvem vil joine os og få disse spændende kandidater videre ud?? Det ser unægteligt meget meget spændende ud.

                                          Anavex Jeg tog sidste år noget gevinst hjem, og genkøbte (desværre for tidligt) efterfølgende. Nu står vi så overfor den mest afgørende periode i selskabets historie. Ved positive resultater i ALZ 2b/3 inden længe vil kursen helt sikkert langt overstige sidste års max lige rundt 30 dollar. Derfor skal vi,i hvert fald jeg selv til at overveje en strategi i.f.t at tage yderligere gevinst. Kommer der gode resultater er risikoen taget ud,og så alligevel, der kan altid komme noget uventet i biotek.
                                          Mener i stadig at de kan undgå opkøb (poison pill intakt?). En partner, som jo er nem at få i dette tilfælde, vil jo i udgangspunktet være af det gode. Det vil dog så være evig slut med at gå alene tænker jeg. Altså vi kommer i mine øjne, over tid, til at dele hele biksen med denne partner hvis Missing ikke er skarp.
