    A new study suggests that an older GLP-1 drug may help protect the brains of people with early Alzheimer's disease, supporting the case for further research on the class of medications - originally developed for obesity and diabetes - in neurological diseases.

    The Phase 2 randomized trial, led by researchers at Imperial College London, tested Novo Nordisk's liraglutide, the predecessor to Ozempic and Wegovy, in patients with early Alzheimer's disease over one year. The study did not meet the primary endpoint of change on a measure of how much sugar the brain uses for energy, but it showed that patients on the drug had nearly 50% less shrinking in parts of the brain that control memory and learning and that treated participants had a slightly slower decline in cognitive function.

        "Novo's GLP-1 drug liraglutide slows cognitive decline by 18% in mid-stage Alzheimer's study

        New research suggests that Novo Nordisk’s GLP-1 agonist liraglutide can protect the brains of people with mild Alzheimer’s disease, slowing cognitive decline by as much as 18% after one year of treatment versus placebo. The Phase IIb study findings were reported Tuesday at the Alzheimer’s Association International Conference (AAIC).

        GLP-1 receptor agonists such as liraglutide – which is currently approved as Saxenda for weight loss and Victoza for diabetes – have been shown in animal models to have neuroprotective effects. The new research presented at AAIC suggests that liraglutide works by slowing the shrinking of the parts of the brain that are vital for memory, learning, language and decision-making".

          Ja der er relativ meget evidens for at GLP1 har en positiv effekt indenfor AD. Og det nye forsøg med liraglutide bekræfter blot dette.

          Liraglutide er første generation GLP1 som ikke er lige så potent som Semaglutide (Ozempic/wegovy/Rybelsus)

          Novo bruger en håndfuld mia på 2 kæmpe store fase 3 forsøg med 14mg Rybelsus. Altså pille udgaven.

          Det bliver ufattelig spændende at se data næste år. MEN jeg kan godt frygte der ikke opnås signifikans. Fordi de kun bruger 14mg vs 25mg og 50mg som novo senere har udført rigtig positive fedme og T2D forsøg med.

          Hvis kommende forsøg med Amycretin viser fin bivirkningsprofil, så tror jeg novo tager en runde 2 med nye AD forsøg.

          Novo har selv sagt at alt det Semaglutid er god til, der er CagriSema endnu bedre. Ud fra deres prækliniske forsøg og herunder ikke mindst inflammationshæmmende som er en af de mekanismer man tror virker på AD.

          Amycretin er jo ét molekyle der både aktivere GLP1 og amylin.Altså i stedet for 2 molekyler som I CagriSema.

          Så HVIS vi ser god bivirkningsprofil og effekt i de 3 igangværende fase 1 forsøg med Amycretin. Så ser jeg Amycretin som deres næste platform hvorfra de kommer til at starte RIGTIG mange fase 3 forsøg de kommende år.
