Johnson & Johnson's mAb looks set to win on all fronts over the next few years in the multiple myeloma market.

Data from Johnson & Johnson's Phase II LYNX study, which due to readout late this year or in early 2023, could provide some answers. This trial is evaluating Darzalex in combination with Kyprolis (carfilzomib) and dexamethasone (Kd), versus Kd alone, in approximately 230 patients that have received one to three prior lines of therapy, one of which contained Darzalex.

Feedback from our poll overwhelmingly supports the assertion that MM patients can be re-treated with Darzalex, on the assumption that there has been an extended break in exposure to the drug. All 30 specialists said they are comfortable re-treating patients with Darzalex in these circumstances, with 50% of respondents describing themselves as 'very' comfortable with this approach and 37% as 'extremely' comfortable.

oncurrently, Darzalex is also being evaluated as a potential maintenance treatment for MM. In the Phase III AURIGA trial, the combination of Darzalex plus Revlimid (lenalidomide) versus Revlimid alone is being studied as maintenance treatment in approximately 214 patients with newly diagnosed MM who are MRD positive following high-dose therapy (HDT) and autologous stem cell transplant (ASCT).

The primary outcome measure is percentage of patients achieving MRD-negative status with results anticipated next year.

A third of the physicians said they are extremely confident Darzalex will become an established maintenance therapy for MM based on its known clinical profile, with a further 53% describing themselves as being very confident of this outcome.

The position of Darzalex as both a key growth driver for Johnson & Johnson and anchor point for its longer-term ambitions in the MM market look set to remain undiminished.
