<?xml version="1.0" encoding="UTF-8"?><rss xmlns:dc="http://purl.org/dc/elements/1.1/" xmlns:content="http://purl.org/rss/1.0/modules/content/" xmlns:atom="http://www.w3.org/2005/Atom" version="2.0"><channel><title><![CDATA[Bioinvent - en baby Genmab?]]></title><description><![CDATA[<p dir="auto">Jeg skal ikke kloge mig på hverken Genmab eller biotech generelt, men blot henlede opmærksomheden på svenske Bioinvent, som er en mulig &quot;baby Genmab&quot; - dvs de gør sig også i antistoffer mod kræftbehandling. Tanken var at få andre til at kigge lidt nærmere på den.</p>
<p dir="auto">De har deres egen teknologi platform, som lige har opnået patent i USA. Det giver dem mulighed for at tjene penge som &quot;underleverandør&quot; af targets for nye antistoffer til de store medicinalfirmaer. Det lægger en hånd under cashburn og emmissioner.</p>
<p dir="auto">Kursmæssigt har Bioinvent levet en omtumlet tilværelse. De havde partneraftaler med store Genetech (blåstempling), men deres førende stoffer fejlede i de kliniske forsøg (ja, det kan ske !) og de er blev slået tilbage til start. Kursen er faldet fra omkring 30 SEK til nu 3-4 SEK.</p>
<p dir="auto">Men de har skåret virksomheden og omkostningerne til og vil nu leve af tidlige partnerskabsaftaler med de store firmaer (har lige indgået ny aftale med Bayer), og de har pt. 3 stoffer enten i start fase 2 eller tæt på klinisk udvikling. Som jeg har forstået det, så er tanken at lave partner aftale på det mest fremskrende stof (BI-505 inden alt for længe)</p>
<p dir="auto">De har også 8 stoffer i tidlig udvikling med 5 partnere og som jeg læser deres seneste kvartalsrapport har de yderligere over 10 forskningsprojekter på vej.</p>

BI-505 is a fully human antibody derived from BioInvent's proprietary n-CoDeR<img

    /> library. The antibody binds to the adhesion protein ICAM-1 (CD54), which is overexpressed in several tumours but not widely expressed in normal tissue. BI-505 is being developed to treat multiple myelomas that express ICAM-1.<br />
Product rationale<br />
BI-505 was identified in a screen to find antibodies that can cause cell death (apoptosis). Antibodies with this property could be used to kill cancer cells to which they bind.<br />
Prior to BioInvent's screen and preclinical studies, the ability of anti-ICAM-1 to lead to apoptosis had not been recognised. Owing to its novel mode of action, BI-505 could be given to patients that do not respond to current treatments.<br />
Clinical status<br />
A dose-escalation phase I study in patients with relapsed or refractory multiple myeloma is on-going. The study explores safety, pharmacokinetics and pharmacodynamics, such as relevant biomarkers for tumour response, with the aim of defining the optimal dose of the antibody for future clinical development. Patients are treated with intravenous doses of BI-505 every second week for a 28-day period with the possibility of extending the treatment until the condition deteriorates.<br />
BI-505 has been granted orphan drug designation for the treatment of multiple myeloma by both the FDA and the European Agency for the Evaluation of Medicines (EMEA).<br />
Development and commercialisation strategy<br />
We are currently developing BI-505 in house at BioInvent.</p>
<p dir="auto">BI-1206<br />
Background<br />
BI-1206 is a so-called antagonistic (blocking) antibody aimed at the immunosuppressive target protein Fc gamma receptor IIB, CD32b. CD32b is overexpressed on tumour cells in patients with lymphoma, especially in patients who respond poorly to currently available drugs. Data show that CD32b is directly involved in the development of tumour cell resistance to the current state-of-the-art treatment -Rituximab (Mabthera<img

    />, Roche), an antibody directed against target protein CD20. Combined treatment with BI-1206 and rituximab has shown significantly enhanced antitumour effects in clinically relevant animal models of patients' tumour cells, compared with monotherapy with rituximab. Combination therapy therefore has the potential to significantly improve treatment of patients with nonHodgkin's lymphoma. BI-1206 has also shown a strong ability to kill lymphoma cells on its own in preclinical models using tumour cells taken directly from patients. Moreover, other groups have shown that animals lacking CD32b (CD32b knockout mice) respond better to antibody treatment and are better able to kill tumour cells in a lung cancer model compared with animals that have the CD32b protein. These results show that BI-1206 also has the potential to be used as monotherapy and that by shutting off the immunosuppressive effect of CD32b and creating a more immunostimulatory environment, it can enhance the therapeutic effect of several previously approved antibody-based drugs other than rituximab. BI-1206 will initially be developed for non-Hodgkin's lymphoma lymphoma (including chronic lymphocytic leukaemia), the most common type of hematologic cancer. Preclinical studies are also planned for assessment of the potential of this antibody to be effective for other types of hematologic cancer, for solid tumours and in combination with antibodies other than rituximab. The product will be developed in cooperation with a leading research group in Southampton, UK. BioInvent believes that market for treatment with BI-1206 combined with other antibodies is significant. In 2012, Rituxan alone had sales of USD 7.1 billion, the majority for hematologic cancer, which makes it the second most sold antibody-based drug worldwide. In various studies, up to half of all cancer patients who responded to an initial course of Rituxan proved to be resistant to the drug on recurrence of the disease.<br />
Project status<br />
Development of the production process for BI-1206 has begun. The next stage of development after up-scaling and production involves toxicological studies, which are expected to begin in early 2014.</p>
<p dir="auto">ADC-1013<br />
Background<br />
ACD-1013 is a so-called agonistic (activating) immunostimulatory antibody developed for local administration into tumour tissue (intratumoural administration). The antibody is directed against the CD40 antigen, which is expressed on several types of immune system cells and stimulation of this protein activates the body's own defence mechanisms against cancer. CD40 is also expressed on several types of tumours, including lymphoma. ADC-1013 and a mouse-specific surrogate antibody have been studied in several different tumour models and consistently shown very promising effects. For example, it has been shown that local administration causes systemic immune activation, resulting in eradication of metastases. In addition, long-lasting immunity against the cancer is also created, thereby protecting against new metastases even after discontinuation of treatment. It has also been shown that the effect can be achieved at lower doses than when administering the antibody systemically, and that the risk of side effects is lower at the same doses. Project status<br />
BioInvent obtained the right to co-develop the product candidate ADC-1013 with Alligator Bioscience through an option agreement. <p dir="auto">Det stora värdet i Q3-rapporten fanns i det förtydligande av det<br />
sammanlagda värdet på 55 miljoner kronor från senaste tidens nya<br />
avtal och uppnådda milstolpar, vilket tidigare inte kommunicerats.<br />
Nivån var klart högre än vi räknat med, cirka 20 miljoner kronor.<br />
Ytterligare preklinisk data för BI-505 kommer presenteras på ASH i<br />
början av december som visar på potential vid kombinationsbehandling<br />
med etablerade läkemedel, vilket är en till pusselbit för att få till<br />
ett licensavtal. Bioinvent-aktien har gått starkt senaste tiden med<br />
hjälp av ett positivt sektorsentiment. Trots bra utveckling för<br />
aktien framstår värderingen fortsatt attraktiv i relation till<br />
sektorn. Vårt motiverade värde för Bioinvent uppgår till 6,7 kronor<br />
(6,2 kronor).</p>
<p dir="auto">Nyhetsvärdet i rapporten för tredje kvartalet kretsar främst kring att<br />
bolaget förtydligar det sammanlagda värdet på de avtal och uppnådda<br />
milstolpar under senaste månaderna, vilket summeras till 55 miljoner<br />
kronor. Detta var klart mer än vi räknat med vilket var cirka 20<br />
miljoner kronor. Vi gissar på att huvuddelen kommer från avtalet med<br />
Bayer som utökar sitt sedan tidigare samarbete kring bolagets<br />
antikroppsbibliotek, nCoDeR. Det verkar</p>
<p dir="auto">dock som att Bioinvent inte kommer få ytterligare milstolpsbetalningar<br />
och royalty från de nya projekten i avtalet, vilket förklarar den<br />
höga kontant ersättningen. Med intäkter på 7,8 miljoner kronor<br />
rapporterat i tredje kvartalet är det ingen vild gissning att vi får<br />
se det mesta från dessa intäkter att utbetalas under innevarande<br />
kvartal. Vi bedömer att intäkter på cirka 50 miljoner kronor hamnar<br />
under innevarande kvartal, vilket tar bolaget till svarta siffror och<br />
positivt kassaflöde under kvartalet och andra halvåret. Det är ett<br />
riktigt välkommet tillskott för bolaget, då likvida medel vid<br />
utgången av tredje kvartalet uppgick till 40 miljoner kronor. Vi<br />
förväntar oss att Bioinvents likvida medel ska överstiga 60 miljoner<br />
kronor vid utgången av 2013. Även om året avlutas starkt på<br />
intäktssidan bedömer vi att bolaget inte når hela vägen till balans<br />
mellan kostnader och intäkter, utan att helårsresultatet trots allt<br />
hamnar på minus. Finansiellt är Bioinvent på en hyfsat stabil nivå,<br />
speciellt om bolaget kan fortsätta leverera på sin målsättning att<br />
med nya avtal kring sina utvecklingsplattformar och att partners<br />
antikroppsprojekt avancerar till kliniska prövningar. När vi talar<br />
med bolaget framstår de relativt konfidenta att ett eller fler<br />
partnerprogram ska kunna avancera till kliniska studier närmaste<br />
månaderna, vilket i så fall aktiverar milstolpsbetalning till<br />
Bioinvent på i storleksordningen 10 miljoner kronor per projekt.<br />
Antalet projekt i preklinisk fas är för närvarande sju och flera av<br />
dessa har varit i preklinisk fas i snart ett år, vilket talar för att<br />
nästa steg inte ligger alltför långt bort i tid.</p>
