Detailed results from Roche's POLARIX study are poised to be one of the highlights at next week's annual meeting of the American Society of Hematology (ASH).

Roche notes that POLARIX is the first positive DLBCL Phase III study in a curative setting for two decades. Furthermore, it has suggested that sales of Polivy could peak at around $2 billion if the antibody-drug conjugate (ADC) becomes established as a new standard of care in the first-line setting.

In response to publication of the abstract, we snap-polled 43 oncologists, 56% of whom said the Polivy-based regimen looks superior to current standard of care based on PFS data alone.

35% says acceptable adverse effect profile and 51% says manageable

Taking efficacy and safety into account, results from POLARIX were described by 44% of oncologists as clinically meaningful and by a further 40% as promising, suggestive that physicians still want to see further detailed results and ideally a survival benefit (despite DLBLC being a relatively slowly progressing disease).

And then they want to see OS data

Regarding Genmabs DuoBody agreement with Janssen we maybe could get an idea of how backloaded the deal is by looking at Zymeworks deal with Janssen.

For 6 programs Zymeworks got 50 million dollars upfront and are eligible for up to $282M in development milestone payments as well as $1.12B in commercial milestones. Thats about 233 milliion dollars pr program which is bit more than Genmabs deal but also 5 years later. So kind of the same I guess.

So that means around 20% of the milestone are development and 80% commercial - so quite backloaded I must say
